thalidomide worked. In a study of the nature, aetiology and pathology of what was named ‘dysmelia’,1-4 Willert and Henkel2,3 arranged a series of 287 thalidomide cases in order of increasing severity of limb reduction. Figures 1 and 2 summarise the pattern of the malformations5 and the teratological sequences which emerged from that study.

To investigate this issue we studied 11 subjects with different degrees of upper extremity dysmelia due to thalidomide embryopathy in comparison to 10 control subjects. In the affected subjects fingers are typically missing in radio-ulnar order beginning with the thumb. Dysmelia thalidomide

Thalidomide is a potent teratogen causing dysmelia (stunted limb growth) in humans. We have demonstrated that orally administered thalidomide is an inhibitor of angiogenesis induced by basic fibroblast growth factor in a rabbit cornea micropocket assay. Experiments including the analysis of thalidomide analogs revealed that the antiangiogenic activity correlated with the teratogenicity but not thalidomide was advertised as a sedative drug to treat anxiety, nausea and morning sickness.1 Maternal intake of this drug between 1958 and 1963 resulted in 8,000–12,000 infants with thalidomide embryopathy (TE), mainly in Europe and Japan.2 Dysmelia is a characteristic feature of thalidomide worked. The pathogenesis of longitudinal reduction deformities of the limbs, or dysmelia, is still a matter of debate. Their morphological pattern was defined from a large collection of radiographs of children with dysmelia following the thalidomide disaster.
